چکیده
Using autologous serum for the serological analysis of recombinantly expressed clones (SEREX) from a cDNA derived from a human mela noma, several new melanoma antigens were identified that are immuno genic in the autologous host. Sequence analysis revealed that one of these antigens, HOM-MEL-40, was coded for by the SSX2 gene, which has recently been described to be involved in the t(X;18) translocation of human synovial sarcomas. Expression analysis performed by Northern blot and RT-PCR demonstrated the presence of HOM-MEL-40 tran scripts in a significant proportion of human melanomas (50%), colon cancers (25%), hepatocarcinomas (30%), and breast carcinoma (20%) but not in normal tissues except for testis. Sequence comparison with tran scripts cloned from testis ruled out mutations in the melanoma-derived HOM-MEL-40. Antibodies against HOM-MEL-40 were found in 10 of 89 patients with melanoma, including 3 of 8 patients with HOM-MEL-40positive tumors, but not in 41 apparently healthy controls. In view of the specific expression pattern and hnmunogeniclty In cancer patients, HOM MEL-MIholds promise as a target for immune interventions in a consid erable population of patients with HOM-MEL-40-positive tumors.

Serological analysis of a recombinant cDNA expression library (SEREX) derived from two lung adenocarcinoma cancer cell lines using autologous sera led to the isolation of 41 positive cDNA clones comprising 28 different antigens. They coded for a variety of nuclear and cytoplasmic proteins. Among the antigens, nucleoporin 107 (NUP107) was isolated most frequently (5 of 41 clones). The second mos...

The identification of human cancer antigens has opened new approaches to the development of cancer vaccines. Characterization of the defined antigens including mRNA expression analysis, protein analysis, antibody response and epitope identification are important to assess the value of those antigens. The aim of the present work is to identify tumor specific or tumor associated antigens by serolo...

The identification of the antigenic stimuli of B-cell neoplasms might be of considerable importance since a causal relationship between these neoplasms and antigenic stimulation has been suggested. To date the identification of such antigens has been erratic and accidental. Cutaneous T-cell lymphomas (CTCL) are a group of skin neoplasms that originate from T lymphocytes and are difficult to treat in advanced stages. The present study is aimed at the identification of tumor-specific antigens from a human testis cDNA library using human sera known as the SEREX (serological identification of recombinantly expressed genes) approach. A cDNA library from normal testicle...
